Source text - Japanese 代謝（たいしゃ、metabolism）とは、生命の維持のために有機体が行う、外界から取り入れた無機物や有機化合物を素材として行う一連の合成や化学反応のことであり、新陳代謝の略称である。これらの経路によって有機体はその成長と生殖を可能にし、その体系を維持している。代謝は大きく異化 (catabolism) と同化 (anabolism) の2つに区分される。異化は物質を分解することによってエネルギーを得る過程であり、例えば細胞呼吸がある。同化はエネルギーを使って物質を合成する過程であり、例えばタンパク質・核酸・多糖・脂質の合成がある。代謝の化学反応は代謝経路によって体系づけられ、1つの化学物質は他の化学物質から酵素によって変換される。酵素は触媒として、熱力学的に不利な反応を有利に進めるため極めて重要な存在である。また、酵素は、細胞の環境もしくは他の細胞からの信号（シグナル伝達）の変化に反応することにより代謝経路の調節も行う。有機体の代謝はその物質の栄養価の高さがどれだけか、また、毒性の高さがどれだけかを決定する。例えば、いくつかの原核生物は硫化水素を使って栄養を得ているが、この気体は動物にとっては毒であることが知られている。また、代謝速度はその有機体がどれだけの食物を必要としているかに影響を与える。代謝の目的は、以下の4つが挙げられる。エネルギーを獲得する。摂取した栄養素を、生体を構成するための前駆体へ転換する。前駆体から生体の成分を合成する。細胞が使う生理活性物質を合成または分解する。

Through these pathways, organisms allow their own growth and reproduction, maintaining their system. Metabolism is largely divided into catabolism and anabolism. Catabolism is a process of obtaining energy by decomposing substances, including cell respiration. Assimilation is a process of synthesizing substances using energy, including synthesis of protein, nucleic acid, polysaccharide and lipid. Metabolic chemical reactions are organized by metabolic pathways and one chemical is enzymatically converted from other chemicals. As a catalyst, enzymes are extremely important for promoting thermodynamically unfavorable reactions advantageously. The enzyme also regulates the metabolic pathway by responding to changes in the cell's environment or signals (signaling) from other cells. The metabolism of the organism determines how high the nutritional value of the substance is and how high the toxicity is. For example, some prokaryotes are nourished by using hydrogen sulfide, whereas this gas is known to be poisonous for animals. Also, the rate of metabolism affects how much food the organism needs. The objectives of metabolism include the following four. To acquire energy. To convert ingested nutrients to precursors for constituting living bodies. 真正細菌における遺伝子の発現調節はジャコブとモノーの研究論文を基礎として理解することができる。発現調節にはオペロンと言う、いくつかの遺伝子が短い間隔を置いて、ゲノム中に並んでいる構造体が深く関与しているが、その発端となった大腸菌のラクトース（乳糖）の代謝について説明する。
ラクトースオペロン
真正細菌及び古細菌では、機能の関連した遺伝子が染色体上で隣接して存在し遺伝子クラスター（gene cluster）を形成している。この遺伝子クラスターのうち、単一のプロモーターで転写される単位をオペロン（operon）という。その代表的なものが、ラクトース（lac）オペロンである。ラクトースは大腸菌の細胞表層から細胞内へ輸送され、その後グルコースとガラクトースに分解される。細胞内への輸送はラクトースパーミアーゼ、グルコースとガラクトースへの分解はβ-ガラクトシダーゼが関与している。この2種類の酵素が同時に働くことによって、大腸菌はラクトース代謝が可能となる。ラクトースオペロン上には、β-ガラクトシドトランスアセチラーゼをコードする遺伝子も存在するが、この酵素はラクトースの資化には直接関係なく、その役割は不明である。β-ガラクトシダーゼ、 β-ガラクトシドトランスアセチラーゼ、ラクトースパーミアーゼはそれぞれ、lacZ、lacA、lacY'という遺伝子によってコードされているが、これらの遺伝子は極めて近接している。ジャコブとモノーはこれらの遺伝子が以下のように配列していることを同定した。
プロモーター(P)-lacZ-lacY'-lacA-ターミネーター(T)
これらの遺伝子群は構造遺伝子と呼ばれており、実際に反応に機能しているタンパク質をコードしている。これらの遺伝子が転写されると、翻訳も同時に進行し、必要なタンパク質全てが発現する。更に、lacZの上流にlacIと言う遺伝子が発見された。この遺伝子は独自のプロモーターおよびターミネーターを持っており、ラクトースの代謝に直接関与するタンパク質をコードしていなかった。lacIの機能は構造遺伝子の転写を調節しており、ラクトースリプレッサーと呼ばれるタンパク質をコードしている。ラクトースの非存在下でこのタンパク質が発現している間は、構造遺伝子の転写は行なわれない。ラクトースリプレッサーは構造遺伝子のプロモーター配列の近傍に存在するオペレーター配列に結合することによって、RNAポリメラーゼの結合を回避させている。逆にラクトースが存在している場合、ラクトースリプレッサーにラクトースが結合し、ラクトースリプレッサーはコンフォメーション変化を起こしてオペレーター配列に結合できなくなる。その時に初めてRNAポリメラーゼが構造遺伝子のプロモーターに結合し、転写が開始される。この反応によってラクトースが消費しつくされると、ラクトースリプレッサーがはたらき転写が抑制される。こうした調節因子（今回はlacI）の働きを変える因子（今回はラクトース）の事をインデューサーという。詳しくはラクトースオペロンを参照。遺伝子の制御に関わる、他の因子としては転写、翻訳の速度やmRNAの回転率などがある。遺伝子の発現に関わる全ての因子がその制御に関わるといってよい。
Translation - English Regulation of gene expression in eubacteria can be understood on the basis of research papers of Jacob and Monod. The regulation of expression deeply involves a structural unit known as operon consisting of several genes which are spaced at short intervals in the genome, and the metabolism of lactose in E. coli that was the origin of this concept shall be explained.
Lactose operon
In eubacteria and archaea, genes with related functions are present adjacent on the chromosome to form gene clusters. Of this gene cluster, the unit transcribed with a single promoter is called operon. The representative one is the lactose (lac) operon. Lactose is transported from the cell cortex of E. coli into the cell, and then decomposed into glucose and galactose. The transport into the cell and the decomposition into glucose and galactose involves lactose permease and β-galactosidase, respectively. The simultaneous action of these two kinds of enzymes allows lactose metabolism of E. coli. Although a gene encoding β-galactoside transacetylase also exists on the lactose operon, the role of this enzyme is unknown due to no direct relationship with the assimilation of lactose. β-galactosidase, β-galactoside transacetylase and lactose permease are encoded by the genes of acZ, lacA and lacY ', respectively, and these genes are very close together. Jacob and Monod have identified that these genes are arranged as follows:
Promoter (P) -lacZ-lacY'-lacA-terminator (T)
These gene clusters are called structural genes, and they encode proteins that actually function in reactions. When these genes are transcribed, translation proceeds simultaneously, and all necessary proteins are expressed. Further, a gene called lacI was found upstream of lacZ. This gene has its own promoter and terminator and did not encode proteins directly involved in lactose metabolism. The function of lacI regulates the transcription of structural genes and encodes a protein called lactose repressor. While this protein is expressed in the absence of lactose, transcription of the structural gene does not occur. The lactose repressor prevents binding of RNA polymerase by binding to the operator sequence located near the promoter sequence of the structural gene. Conversely, when lactose is present, lactose binds to the lactose repressor and the lactose repressor undergoes a conformational change, making it impossible to bind to the operator sequence. At that time, RNA polymerase binds to the promoter of the structural gene for the first time and transcription is initiated. Once lactose is fully consumed by this reaction, the lactose repressor works to inhibit transcription. A factor (lactose in this case) that alters the function of this regulatory factor (lacI in this time) is called an inducer. See lactose operon section for details. Other factors involved in gene regulation include transcription and translation rates and mRNA turnover rates. It can be said that all factors related to gene expression are involved in its control.
